Preparation and Evaluation of Newcastle Disease Oil Emulsion Vaccine at Hydrophile Lipophile Balance 7.0 Using Mukteswar Strain

چکیده
Mukteswar strain of Newcastle disease (ND) virus was propagated in 9-day-old embryonated eggs via allantoic route. The harvested allantoic fluid with haemagglutination (HA) titre of 1024 (EID50 10) was inactivated using 0.1 percent formalin solution. Two experimental vaccines were prepared in light mineral oil with aqueous to oil ratio of 1:2 ( MK-VAC-I) and 1:4(MK-VAC-II).The hydrophile lipophile balance (HLB) of the vaccine was fixed at 7,0 using oil phase (span 80) and aqueous phase (tween 80) surfactants. One hundred and twenty, four week old layer chicken were divided into four equal groups (A to D). Groups A and B were injected with two experimental vaccines and C with commercial oil emulsion vaccine keeping D group as non-vaccinated control. Highest four week cumulative haemagglutination inhibition titres were shown by commercial vaccine (226) followed MK-VAC-I (217) MK-VAC-II (171) and lowest by control group. Both the vaccine showed 80 percent protection against virulent virus challenge comparable to the commercial one.

Two oil emuisified vaccines of Newcaslte disease were prepared from Lasota strain. The virus was propagated in 9-day-old chicken embryos through allantoic route. The haemagglutination titre of the allantoic fluid used for vaccine preparation was 2048. The fluid was inactivated by 0.1 per cent f nal concentration of formalin at 37 oc for 6 hours. Inactivated allantoic fluid was emulsified with a...
متن کاملEvaluation of Quality of Two Commercial Oil-Emulsion Newcastle Disease Vaccines In-vivo and In-vitro
Background and Aims: Newcastle disease (ND) is a highly contagious disease in poultry with economic losses in the world. Vaccination is one of the most important ways for prevention and control of NDV, but there are reports of ND outbreaks in vaccinated chickens. Poor quality of the vaccines is one of the reasons of vaccine failure. In this study the quality of two commercial oil-emulsion Newca...

The present study was conducted to prepare oil emulsified (OE) infectious bronchitis (IB) experimental vaccines. The vaccines were prepared using the vaccinal strain H-120 Infectious Bonchitis virus (IBV). The virus was cultivated in 9-day old embryonated eggs via allantoic cavity route. Allantoicamniotic fluid (AAF) was collected and inactivated with formalin @ 0.12%. Water in oil emulsion was...
متن کاملFormation, Characterization and Stability of Nanoemulsions Prepared by Phase Inversion
Formation and stability of n-undecane in water nanoemulsions prepared by EIP method (Emulsion Inversion Point) in the presence of two nonionic surfactants were studied. In the study, the influences of surfactant concentration, HLB value and oil–to–water ratio on the particle size and distribution were investigated. The stability of emulsions was evaluated from changes in particle size distribut...
